Merrifield Resin Peptide Synthesis - Omizzur

Merrifield resin is a material used for solid-phase peptide synthesis and purification. It is very important for the success rate of peptide synthesis. Different resins have different properties and functions and are suitable for different types of synthesis.


Solid Phase Peptide Synthesis 


There are two main methods used in solid-phase peptide synthesis today: BOC and FMOC. During the synthesis process of the BOC method, TFA must be frequently used to remove BOC, and HF must be used for subsequent cutting from the resin. The FMOC method has stable reaction and can be made directly under normal conditions.


Important parameters about the resin: degree of cross-linking, mesh number, degree of substitution, degree of swelling in different solvents.


1. Cross-linking degree: Taking PS resin as an example, its resin balls are formed by adding a certain amount of cross-linking agent divinylbenzene to polystyrene and undergoing a polymerization reaction. 

The added amount of cross-linking agent divinylbenzene is the degree of cross-linking (DVB). The optimal cross-linking degree of the resin is 1%. A higher degree of cross-linking will lead to a decrease in resin swelling, but a lower degree of cross-linking will reduce the swelling degree of the resin. Mechanical stability of the resin in the swollen state. Generally speaking, most of the resins on the market have a cross-linking degree of 1%.


2. Mesh size: The size of the resin balls. Generally, commercially available resins are 100-200 mesh (average diameter is 75-150 μm). The conditions of the polymerization reaction can affect the size of the resin balls.


3. Degree of substitution: How many millimoles of active reaction sites are loaded per gram of resin, the unit is mmol/g. It is worth noting that after obtaining the commercially available Wang-resin and CTC-resin, we will load the first amino acid in the laboratory, but the amino acid loading rate of these two resins cannot reach 100%, so it should be restarted after that. Determine the degree of substitution.


4. Swelling degree: The swelling ability of resin in different solvents, the unit is ml/g. Generally, after obtaining commercially available resin, we need to use a solvent to swell it before use. 

Good swelling can increase the chance of reactants entering the site. DCM, DMF, and N-methylpyrrolidone are all commonly used benign resin swelling solvents.

There are several factors to consider when selecting a resin. The first is the characteristics of the peptide, such as length, sequence and side chain properties. Different peptides may have different requirements for resin selection. 


Next are the synthesis conditions and purification methods. Some resins perform better in specific solvents and conditions. While others are more suitable for the purification of peptides.
