Protected amino acids are the most basic raw materials for solid-phase peptide synthesis . Amino acids all contain alpha amino and carboxyl groups, and some of the 20 amino acids also contain side-chain active groups, such as hydroxyl, amino, guanidino and heterocyclic groups. These active groups need to be protected during the peptide reaction, and the protective groups are removed after the target peptide is synthesized.The most frequently used protected amino acid raw material is the Fmoc amino acids



Optimization of Fmoc Protected Amino acids Synthesis

In peptide synthesis, in order to obtain the ideal target peptide, it is first necessary to block or protect the active groups of amino acids. Boc and Fmoc are the preferred protective groups in solid-phase synthesis, forming two major categories of peptide solid-phase synthesis methods: boc and Fmoc method.


In the synthesis of Fmoc-Arg(Pbf)-OH, due to some unique advantages of the protective group of Fmoc, such as its instability towards bases and ease of detection, we chose Fmoc as its protective group. In the process of fmoc strategy synthesis, triphenylmethyl (trt) is stable for condensation and deprotection conditions. It can be removed by dilute acetic acid at slightly higher temperature conditions or TFA at room temperature. Therefore, Trt is chosen to block its active functional group. The experiment optimized the two-step protection reaction conditions of His using the central surface experimental design method and orthogonal experimental design method, resulting in a final yield of over 80% for both reactions and a product purity of over 96%.
