Peptides can be obtained through various pathways such as chemical synthesis, biotechnology, protein hydrolysis, and extraction from plant and microbial resources. In the development of peptide drugs, chemical synthesis methods have shown unique advantages.


The full chemical synthesis of peptides can be divided into liquid phase and solid phase synthesis. Since its establishment by Merrifield in 1963, the Solid Phase Peptide Synthesis (SPPS) method has been increasingly used in the research and development of bioactive peptides due to its advantages of simple operation, fewer by-products, and high purification efficiency, especially in the actual production of medicinal short peptides.


By using techniques such as solid-phase synthesis, liquid-phase synthesis, and solid-phase liquid-phase synthesis, peptide chemical synthesis is carried out through amino acid substitution, side chain and end group modification, combinatorial chemistry, and other methods to obtain analogues and derivatives of bioactive peptides. Subsequently, the screening and structure-activity relationship study of highly active peptides are carried out, effectively improving the efficiency of peptide drug development.

In the solid-phase synthesis of peptides, 2-Chlorotriphenyl Chloride Resin (2-CTC) has been applied in the industrial production of multiple peptide drugs such as cell fusion enzyme inhibitory peptides T20 and T1249 due to its milder reaction conditions, effective inhibition of racemization, and high product purity compared to other resins.


The 2-Chlorotrityl Chloride Resin is one of the most frequently used and most versatile resins for the large-scale manufacture of peptides. It is slightly less acid-labile than Trityl Resin and thus more widely used in solid phase peptide synthesis. Peptides UBI-8 and UBI-9 were synthesized by Fmoc-solid phase chemistry using a 2-chlorotrityl chloride linker-resin. 


In addition, 2-Chlorotrityl Chloride Resin can also be used for solid-phase synthesis of carboxylic acids, alcohols, and thiols. Its widespread use and excellent performance determine that it is no longer just a commonly used synthetic carrier in the laboratory, but will gradually become a carrier resin for mass production.


Trityl linkers have certain advantages in comparison to the alkoxybenzyl alcohol (eg. Wang resin), especially in the area of peptide chemistry. First, the problem of racemization during the loading of the first amino acid (using DIPEA) is avoided with trityl linkers. Second, due to steric factors, no diketopiperazine formation is observed.


2-Chlorotrityl Chloride Resin is used in convergent peptide synthesis. Extremely acid labile resin for preparing peptide acids and partially protected peptide fragments by the Fmoc strategy. Cleavage can be effected by using AcOH/TFE/DCM or 0.5% TFA.


2-Chlorotrityl chloride is also used as an intermediate of clotrimazole. It is also used in the preparation of modified trityl nucleosides as inhibitors of P. falciparum dUTPase. In addition, it serves as a protecting group reagent for carboxylic acids forming esters.
