General Description
Kisspeptin-10 is a peptide hormone, which was discovered by independent research in the fields of cancer biology and reproductive endocrinology. It is encoded by KISS1 gene, which was first identified in the study of human malignant melanoma in the mid-1990s. The researchers observed that the expression of this gene inhibited the metastatic ability of cancer cells, so they initially characterized it as a metastasis suppressor gene. Later, the peptide product produced by KISS1 gene was named kisspeptin-10, which reflected the source of the gene and its peptide essence.
Subsequent studies have shown that kisspeptin-10 exerts its biological effects by binding to a specific G protein-coupled receptor, which was originally called GPR54 and later named KISS1R. The identification of this receptor marks a breakthrough in people's understanding of the physiological function of kisspeptin. Genetic studies on human and animal models show that the functional deletion mutation of KISS1R gene will lead to abnormal puberty development, thus establishing kisspeptin's position as a key regulator of reproductive axis. These findings strongly indicate that kisspeptin is at the core of reproductive neuroendocrine regulation.
Kisspeptin-10 was synthesized as a large precursor protein, and then hydrolyzed by protease into several active peptides with different lengths, including those commonly known as kisspeptin-54, kisspeptin-14, kisspeptin-13 and kisspeptin-10. Despite their different lengths, these peptides all share a common C-terminal sequence, which is essential for receptor activation. These peptides are mainly produced in specific areas of hypothalamus and act upstream of gonadotropin-releasing hormone neurons.
The main physiological function of Kisspeptin is to stimulate the secretion of gonadotropin-releasing hormone, and then trigger the release of luteinizing hormone and follicle stimulating hormone from the anterior pituitary. Through this mechanism, kisspeptin controls the onset of puberty, regulates gonadal function, and participates in the coordination of reproductive cycle. Experiments show that injecting kisspeptin into human and animals can significantly increase the level of luteinizing hormone, which proves its powerful stimulating effect on reproductive endocrine system.

Besides its central role in reproduction, other physiological and pathological functions of kisspeptin have also been studied. It was originally identified as a metastasis suppressor gene, which prompted researchers to study its role in cancer biology.
In many cancer types, the decrease of KISS1 expression is related to the increase of metastasis potential, and the restoration of kisspeptin signal transduction can inhibit cell migration and invasion in experimental models. These observations show that kisspeptin signal transduction not only affects endocrine regulation, but also affects cell behavior.
Kisspeptin-10 is also involved in metabolic regulation, placental biology and cardiovascular function, although the research in these fields is not as in-depth as its role in reproduction. During pregnancy, placental tissue will produce a large number of kisspeptin, which is considered to be helpful to the invasion control of trophoblast cells and placental development. During normal pregnancy, circulating kisspeptin level increased significantly, which highlighted its potential as a biomarker for obstetric research.
In clinical and biomedical research, kisspeptin has become an important tool and therapeutic candidate. The synthesized kisspeptin polypeptide was used in experiments to evaluate the function of hypothalamus-pituitary-gonad axis, and to study puberty and fertility disorders.
At present, people are interested in developing kisspeptin-based therapies to treat hypogonadism, infertility and possibly hormone-dependent diseases. Therefore, the discovery of kisspeptin has a far-reaching impact on both basic science and applied medical research, linking molecular genetics, endocrinology and clinical research.
