Peptide synthesis method

Peptide solid - phase synthesis is the most popular method of peptide synthesis. The first amino acid at the C-terminal is coupled to an activated carrier such as polystyrene. The resin can be used as a C-terminal protection group and can be used to rapidly separate the growing peptide product from the reaction mixture formed during synthesis.

The catalytic active sites and most receptor binding sites of the ligands are located on the surface of the domain and subunit. Long peptides are often used to study the binding of proteins to receptors. For example, the figure shows the presence of two active sites by binding receptors (yellow). Both sites are located at the interface between the two subunits of the protein.

Advantages of solid phase synthesis of peptide

Chemosynthesis of proteins of 100 to 200 amino acids is faster and cheaper than other methods, and the chemosynthesis of proteins follows the sequence of amino acids without errors or mutations. Chemical synthesis can also add modifications that cannot be made otherwise, such as replacing regular amino acids with type D amino acids and increasing phosphorylation or methylation at a particular site.Chemical synthesis does not require additional labels such as 6HIS or FLAG for purified proteins. In terms of protein expression methods, gene synthesis, plasmid construction, protein expression and purification occupy the vast majority of time and cost. Therefore, the chemical synthesis method of long peptide is worth trying for the majority of scientific research.

Clients often inquire about the synthesis of very long peptides, such as long peptides with lengths of 100 to 200 amino acids, which are already classified as proteins. Most orders are for peptides less than 50 amino acids, but through our modification of peptide linking technology and innovation for long peptides, we can now successfully synthesize peptides of protein length. At present, the longest peptide synthesized by us is 151 amino acids, which proves that our synthesis technology is fully competent for the synthesis of long peptides in the existing technology platform.
