Peptide Synthesis Cost - Omizzur Peptide Services

Peptide is a biomolecule with amino acid as the basic unit and connected by peptide bond, and its molecular weight is generally less than 10 KDa.
Amino acids are a kind of organic compounds containing both amino and carboxyl functional groups. There are 20 kinds of common amino acids in nature. A typical amino acid has a central α carbon atom, and its connecting groups include amino group (NH2), carboxyl group (COOH), hydrogen atom, and variable side chain (R group) that determines the characteristics of amino acids.
When multiple amino acids are dehydrated and condensed with the amino group of the next amino acid through the carboxyl group of the previous amino acid to form peptide bonds, peptide chains are formed.
Solid phase peptide synthesis is an important breakthrough in peptide synthesis chemistry. Its main feature is that there is no need to purify intermediate products, and the synthesis process can be carried out continuously, which lays the foundation for the automation of peptide synthesis of Omizzur peptide. At present, the automatic synthesis of peptides is basically solid-phase synthesis. The basic process is as follows:

FAQs of Custom Peptide Synthesis
1. How to choose the purity of peptide in different experiments?
The purity of peptide is a very important index, and the choice of purity depends on the purpose of the experiment. For screening tests with low sensitivity, it is recommended to use crude products or > 75%, and for immunity level, it is recommended to use > 85%.
For receptor-ligand interaction research, bioassay or cell level research, the recommended value is > 95%, and for structure research, the recommended value is > 98%.
2. What is the net content of peptide (peptide content) and what is its significance?
The weight of dry peptide not only contains peptide, but also contains some non-peptide components, such as water, absorbed solvents, coordination ions and salts. The net content of peptide refers to the weight percentage of peptide. The value of this percentage can be in the range of 50% to 90%, depending on purity, sequence and synthesis and purification. Don't confuse the net content of peptide with the purity of peptide. They are two completely different concepts.
The purity is usually determined by HPLC. Purity is defined as the percentage of components with correct sequence in the peptide sample, while the net content of peptide refers to the percentage of peptide substances relative to non-peptide substances in the sample. The net content of peptide is usually determined by amino acid composition analysis or ultraviolet spectrophotometry.
In some experiments sensitive to peptide concentration, this information is very important for calculating peptide concentration.
3. How long peptide segment is suitable for synthesis?
Peptide synthesis needs to consider the length, charge and hydrophobicity of the peptide. The longer the length, the lower the purity and yield of crude products, the more difficult the purification, and the greater the probability of synthesis failure. Of course, the sequence of polypeptide functional region cannot be changed, but for the smooth synthesis of polypeptide, it is sometimes necessary to add some auxiliary amino acids in the upstream and downstream of functional region to improve the solubility and hydrophilicity of peptide.
Peptides less than 5 peptides generally need hydrophobic amino acids, otherwise it is difficult to post-process.
4. How to determine the solubility of peptide from peptide sequence?
If the peptide contains a high proportion of hydrophobic amino acids, such as Leu, Val, IIe, Met, Phe and Trp, it is difficult to dissolve in aqueous solution or not at all. These amino acids, whether purified or synthesized, may be problematic.
Generally, the proportion of hydrophobic amino acids is less than 50%, and five consecutive amino acids cannot be hydrophobic. The proportion of charged amino acids (positive K, R, H and N terminals, negative D, E and C terminals) reaches 20%. In peptide N or C, if polar amino acids can be added, the solubility can also be improved.
5. How to store synthetic peptides?
Long-term preservation of peptides generally requires dark storage, while short-term preservation should be at -20℃ and 4℃. Peptides can be transported at room temperature for a short time. Peptides are stable at -20℃, especially freeze-dried and stored in dryers.
Freeze-dried peptides can be stored at room temperature before exposure to air. This will reduce the influence of humidity. When freeze-drying is impossible, the best way is to store a small amount of samples. For peptides containing Cys, Met or Trp, deoxygenated buffer is essential for their dissolution, because these peptides are easily oxidized by air. Nitrogen or argon flowing slowly through the peptide before capping can also reduce oxidation.
Peptides containing Gln or Asn are also easy to degrade. Compared with peptides without these problems and simple peptides, all these peptides have limited life.
