The chemical synthesis method has the advantages of short development cycle and rapid production. It can be divided into liquid phase and solid phase method. The main difference between the two comes from whether a solid phase carrier is used. Liquid-phase synthesis developed earlier, and solid-phase synthesis is now the main method for peptide synthesis.


Basic Principles of Solid Phase Synthesis:

1) First, connect the carboxyl group of the C-terminal amino acid of the target peptide chain to be synthesized to an insoluble polymer resin in the form of a covalent bond.


2) Using the amino group of the amino acid as the starting point, connect the deprotected N-terminus of the amino acid to the second amino acid whose carboxyl group has been activated through a condensation.


3) Repeat the condensation → washing → deprotection → neutralization and washing → next round of condensation to reach the length of the peptide chain to be synthesized. followed by peptide cutting, modification, separation and purification, finally will obtain the target peptide.


The solid-phase method is suitable for the synthesis of 10-50 amino acids. According to different α-amino protecting groups, solid-phase synthesis methods can be divided into Boc method and Fmoc method. The solid-phase method is easy to operate, it is easy to automate repeated operations, and the product yield and purity are high. It greatly promotes the R&D of peptide drugs.
